Abstract
Background and Objectives
Human neutrophil antigens include 5 antigen systems. These antigens are on the plasma membrane of white blood cells. The aim of the study is to know about HNA allele frequency in the population in order to estimate the prevalence of the diseases related to HNA-5.
Materials and Methods
In this descriptive study, blood samples were taken from 190 Azari unrelated blood donors having referred to Tabriz Blood Center. DNA extraction was performed using commercial kit Pars Tous. HNA-5a and HNA-5b molecular were then evaluated by PCR-RFLP. PCR products were digested using enzymes Sdu1 Bsp1286I. HNA-5a+ and HNA-5a- (or HNA-5b) antigen gene frequencies were calculated using the Hardy-Weinberg equation.
Results
HNA-5a (+/+), HNA-5a (+/-) and HNA-5a (-/-) phenotypes were observed in 93, 10 and 87 patients, respectively. The observed frequencies were consistent with Hardy-Weinberg equilibrium; allele frequencies of HNA-5a+ and HNA-5a- (or HNA-5b) were equal to 0.51 and 0.49, respectively.
Conclusions
HNA-5 alleles frequency rates were in the same range reported for the other populations.
