چکیده
سابقه و هدف 
اگزوزوم‌ها دسته‌ای از وزیکول‌های خارج سلولی هستند که از طریق مسیر اندوزومال آزاد می‌شوند و قطری بین 150-30 نانومتر دارند. محتوای آن‌ها شامل ماکرومولکول‌های زیستی از جمله DNA ، RNA و پروتئین‌ها می‌باشد. اگزوزوم‌های مشتق از پلاکت، فراوان‌ترین نانوذرات جریان خون هستند. از آن جا که یافتن روشی ساده و کارآمد برای جداسازی این وزیکول‌ها بسیار با اهمیت است، در این پژوهش روش مؤثری بر مبنای رسوب پلیمری جهت جداسازی اگزوزوم‌های پلاکتی معرفی شده است. 
مواد و روش‌ها
در این مطالعه تجربی، نمونه‌های کنسانتره پلاکتی تهیه شده به روش پلاسمای غنی از پلاکت پس از یک روز ذخیره‌سازی، از سازمان انتقال خون ایران تهیه شد. در پایان روز 3، دو دستورالعمل مختلف در دورهای سانتریفیوژ و بر مبنای رسوب پلیمری، جهت جداسازی اگزوزوم‌های پلاکتی استفاده شد. اندازه ذرات جدا شده توسط تفرق دینامیکی نور یا DLS مقایسه شد (n=2). هم‌چنین غلظت ذرات جدا شده با روش بردفورد تعیین شد (n=2) و آنالیز وسترن بلات برای نشانگر اگزوزومی CD63 انجام شد.
یافته‌ها
نتایج حاصل از تحلیل DLS نشان داد که در دستورالعمل A، ذرات جداشده دارای توزیع اندازه گسترده‌تری بودند و بیش از 73% ذرات در محدوده خارج از اندازه اگزوزوم (بزرگتر از 150 نانومتر) قرار داشتند. در مقابل، در دستورالعمل B که شامل مراحل سانتریفیوژ در چند دور متوالی و رو به افزایش بود، 100% ذرات در بازه اندازه 30 تا 150 نانومتر (محدوده اگزوزوم) قرار گرفتند و توزیع اندازه بسیار یکنواخت‌تری مشاهده شد. هم‌چنین، نتایج روش بردفورد نشان‌دهنده میانگین غلظت پروتئین ذرات استخراج شده، معادل mg/mL 0.424 ± 0.12 در دستورالعمل A و mg/mL 0.19 ± 2.62 در دستورالعمل B بود. تجزیه و تحلیل وسترن بلات نیز بیان پروتئین  نشانگر اگزوزومی CD63 را در ذرات جداشده تأیید کرد. 
نتیجه گیری            
در این مطالعه مشخص شد که انجام چند مرحله سانتریفیوژ متوالی در روش رسوب پلیمری، نقش مؤثری در جداسازی اگزوزوم‌های خالص‌تر با اندازه یکنواخت دارد. این مطالعه روش بهینه شده‌ای را جهت جداسازی اگزوزوم‌های خالص‌تر بر مبنای رسوب پلیمری معرفی می‌کند.
