<p> <strong> میکروپارتیکلهای مشتق از پلاکت در دو محیط مختلف پلاسما و کمپوسول </strong></p><p> </p><p> </p><p> <strong><i> شیما آزادپور<sup>1</sup>، فاطمه یاری<sup>2</sup>، شهرام وائلی<sup>3</sup> </i></strong></p><p> <strong> </strong></p><p> چکید ه </p><p> <strong><i> سابقه و هدف </i></strong></p><p> <strong> پلاکتهای فعال شده در پاسخ به محرکهای خاص، میکروپارتیکلها را در بدن و یا با گذشت زمان، در طول ذخیره در کیسه پلاکتی، آزاد مینمایند. هدف از این مطالعه، مقایسه تاثیر دو محیط ذخیره متفاوت شامل پلاسما و محلول افزودنی(کمپوسول) بر تولید میکروپارتیکلهای مشتق از پلاکت در طول زمان نگهداری بود. </strong></p><p> <strong><i> مواد و روشها </i></strong></p><p> <strong> در یک مطالعه تجربی، 30 کیسه کنسانتره پلاکتی از بانک خون ایران تهیه و به دو بخش با حجم یکسان تقسیم شد و سپس پلاسمای یکی از این بخشها با محلول افزودنی کمپوسول جایگزین گردید. از هردو نوع کیسه در روزهای 2، 4 و 7 ذخیره، نمونهبرداری شد. میکروپارتیکلهای مشتق از پلاکت جدا شده و جهت تعیین غلظت آنها از روش اندازهگیری غلظت پروتئینی برادفورد استفاده شد. نتایج با استفاده از آزمون آماری </strong><strong>t </strong><strong>تجزیه و تحلیل شدند. </strong></p><p> <strong><i> یافتهها </i></strong></p><p> <strong> تولید میکروپارتیکلهای پلاکتی در دو محیط نگهداری مختلف، در طول زمان ذخیره افزایش مییابد. تفاوت غلظت نهایی میکروپارتیکلهای مشتق از پلاکت در فرآورده کنسانتره پلاکتی، در روز 4 ذخیره در دو محیط پلاسما وکمپوسول معنادار نبود در حالی که در روز 7 ذخیره این تفاوت معنادار شد و در پلاسما بیش از کمپوسول گردید(05/0 </strong><strong>p< </strong><strong>). </strong></p><p> <strong><i> نتیجه گیری </i></strong></p><p> <strong> در مطالعه حاضر، در میزان تولید میکروپارتیکلهای مشتق از پلاکت در دو محیط مختلف نگهداری(پلاسما و کمپوسول)، تفاوت معناداری وجود نداشت. هر چند پس از 7 روز این تفاوت معنادار گردید و برای پلاسما نسبت به کمپوسول افزایش نشان داد که خود میتواند یک مزیت برای محیط نگهداری کمپوسول از این جهت محسوب گردد. </strong></p><p> </p>
<p> <strong> Abstract </strong><strong></strong></p><p> <strong><i>Background and Objectives</i></strong></p><p> Activated platelets release microparticles (MPs) <i>in vivo</i> or in the platelet concentrate (PC) product in response to some stimulators. This study compared the two different storage media of plasma and Composol for PC regarding the extent of MPs formation as a new index for comparing platelet stability. </p><p> </p><p> <strong><i>Materials and Methods</i></strong></p><p> In this experimental study, 30 PC units were prepared. The platelets were divided into two equal portions. Plasma was replaced with the additive solution of Composol in one of the portions. Sampling was carried out at the days 2, 4 and 7 after storage. Afterwards, the MPs were separated and their concentrations were collected using Bradford method. T-tests was used to compare the results of this experiment. </p><p> </p><p> <strong><i>Results</i></strong></p><p> The results showed that the amounts of MPs increased during the storage. In each media of plasma or Composol, the concentration of MPs showed a significant difference between the days 4 and 7 of the storage. Besides, the final concentration of MPs did not show a significant difference between the two media at the day 4 of the storage whereas this difference was significant at the day 7 of storage (p<0.05). </p><p> <strong><i> </i></strong></p><p> <strong><i>Conclusions</i></strong></p><p> There were no significant differences in the quantity of MPs in PCs stored in plasma or Composol up to the day 4 of the storage. This difference became significant after 7 days of the storage, in which the generation of MPs showed significant increase in plasma than Composol. These observations revealed an advantage for Composol as a PC storage medium. </p><p> </p><p> </p>
