سابقه و هدف&amp;nbsp;&lt;br&gt;
در طول عفونت مزمن ویروس هپاتیت B ، تعادل پویا بین تکثیر ویروس و پاسخ ایمنی میزبان در ایجاد بیماری کبدی از اهمیت زیادی برخوردار است. اعتقاد بر این است که پاسخ&#8204;های ایمنی اکتسابی، به ویژه پاسخ&#8204;های ایمنی سلولی، منجر به پاکسازی HBV می&#8204;شود. PD-1، نقش منفی در تنظیم فعال&#8204;سازی و تکثیر سلول&#8204;های T و تولید سیتوکین&#8204;ها دارد و با مهار عملکرد سلول&#8204;های CD8+ T اختصاصی ویروس، در عفونت مزمن ویروس هپاتیت (CHBV) نقش دارد. در این مطالعه، میزان بیان مرگ برنامه&#8204;&#8204;ریزی شده نوع 1 در اهداکنندگان فاقد علائم مبتلا به هپاتیت B طی سیر طبیعی عفونت ویروس هپاتیت B بررسی شد.&amp;nbsp;&lt;br&gt;
مواد و روش&#8204;ها&lt;br&gt;
در این مطالعه مورد-شاهدی، 120 اهداکننده خون به دو گروه تقسیم شدند. گروه کنترل شامل 60 فرد سالم HBsAg منفی، و گروه مورد شامل 60 اهداکننده خون بدون علامت آلوده به HBV و HBsAg مثبت بود. بر روی نمونه&#8204;های HBsAg مثبت، آزمایش&#8204;های anti-HBc تام، HBeAg و anti-HBe، آلانین آمینوترانسفراز (ALT) و آسپارتات آمینوترانسفراز (AST) &amp;nbsp;انجام شد. برای بررسی بیان ژن پس از استخراج RNA و ساخت cDNA ، آزمایش Real-Time PCR برای اندازه&#8204;گیری بیان ژن PD-1 و ژن مرجع &amp;beta;-اکتین انجام شد. تغییرات در بیان ژن با استفاده از نرم&#8204;افزارREST، با آستانه معناداری 05/0p&lt; تجزیه و تحلیل شد.&lt;br&gt;
یافته&#8204;ها&lt;br&gt;
تمام نمونه&#8204;های اهداکنندگان فاقد علایم مبتلا به هپاتیت B از نظر Total anti-HBc وanti-HBe &amp;nbsp;، مثبت و از نظر HBe Ag منفی بودند. سطح آنزیم های &amp;nbsp;ALT &amp;nbsp; و AST برحسب IU/L به ترتیب برابر 11/0 &amp;plusmn; 29 و 21/0 &amp;plusmn; 28بود . میزان بیان ژن با استفاده از نرم&#8204;فزار REST و ctهای به دست آمده از آزمایش Real-Time PCR برای ژن PD-1 و ژن رفرانس تجزیه و تحلیل شد و افزایش 5/1 برابر بیان ژن PD-1 در اهداکنندگان فاقد علائم مبتلا به هپاتیت B نسبت به افراد سالم نشان داده شد که از نظر آماری معنادار نبود.&lt;br&gt;
نتیجه گیری&amp;nbsp;&amp;nbsp; &amp;nbsp;&amp;nbsp;&amp;nbsp; &amp;nbsp;&amp;nbsp;&amp;nbsp; &amp;nbsp;&lt;br&gt;
بیان ژن PD-1 در طول سیر طبیعی عفونت مزمن HBV در افراد فاقد علایم مبتلا به هپاتیت B به طور معنادار تغییری نمی&#8204;کند و مشابه افراد سالم است.&lt;br&gt;
