fa فراوانی گروه‌های آللی کمیاب HLA-A در قومیت‌های گیلک، لر، کرد و عرب ایران Frequency of rare HLA-A allelic groups in Gilak, Lor, Kurdish and Arab ethnic groups of Iran ايمونولوژي Imunology پژوهشي Research <div style="text-align: justify;">چکیده سابقه و هدف آنتی‌ژن‌های سازگاری نسجی(HLA)، تنوع ژنتیکی بالایی داشته و فراوانی آنها می‌تواند در قومیت‌های مختلف، متفاوت باشد. لذا تعیین این آنتی‌ژن‌ها در قومیت‌های مختلف می‌تواند داده‌هایی به دست دهد که از آن بتوان در گسترش مرکز ملی پذیره‌نویسی اهداکنندگان سلول‌های بنیادی استفاده نمود. مواد و روش‌ها در این مطالعه توصیفی که در طی سال‌های 1399 و 1400 بر روی 2064 داوطلب اهدای سلول‌های بنیادی خونساز صورت گرفت، تعیین آلل‌های HLA-A با روش PCR-SSP صورت پذیرفت. اطلاعات نژادی/قومیتی و HLA از قومیت‌های گیلک(510 نفر)، لر(465 نفر)، کرد(719 نفر) و عرب(370 نفر) تجزیه و تحلیل و ارتباط آلل و نژاد با روش آماری کای‌دو، ارزیابی شد. برای این منظور، مقایر باقی‌مانده استاندارد شده، تعیین گردیده و در تجزیه و تحلیل داده‌ها از نرم‌افزار R استفاده شد. یافته‌ها فراوانی آللی در گروه‌های قومی مورد مطالعه، برای HLA-A*23 و HLA-A*31 ، کمتر از 3 درصد به دست آمد. در گروه‌های آللی HLA-A*25 ، HLA-A*34 ، HLA-A*36 ، HLA-A*43 ، HLA-A*66 ، HLA-A*69 و HLA-A*74 ، این فراوانی کمتر از 1% جمعیت مورد مطالعه بود. فراوانی گروه‌های آللی کمیاب HLA-A ، شامل HLA-A*34 ، HLA-A*69 و HLA-A*74 ، در اقوام لر، گیلک، عرب و کرد تفاوت معنادار(p- value کمتر از 05/0) نشان دادند. نتیجه گیری شناسایی فراوانی آلل‌های HLA در قومیت‌های مختلف، می‌تواند شباهت‌ها و تفاوت‌ها در گروه‌های آللی را در آن اقوام مشخص و در طراحی یک برنامه بهتر برای توسعه مراکز اهدای سلول های بنیادی مؤثر باشد.  </div> <div dir="rtl" style="text-align: left;">Abstract Background and Objectives Histocompatibility antigens (HLAs) have high genetic diversity and their frequency can be diverse in different ethnicities. Therefore, the determination of these antigens in different ethnicities can provide data used in the expansion of centers for the registration of stem cell donors. Materials and Methods In this descriptive study, donor DNA was extracted and qualified. Then, the allele groups of HLA-A were determined at low resolution level by PCR-SSP method. Ethnic and HLA information of Gilak (n= 510), Lor (n= 465), Kurdish (n = 719) and Arab (n = 370) were analyzed and the relationship between the allele and ethnicity was examined and a significant relationship between the frequencies of alleles in the studied ethnicities were evaluated by chi-square statistical method. In this study, a statistically significant level of 0.05 was considered. In order to determine in which of the ethnicities the frequency of the studied alleles was higher than the expected values under the assumption of independence, the standardized residual values were determined. Results Allelic frequencies in the studied ethnic groups were obtained for HLA-A*23 and HLA-A*31, <3% . In allelic groups of HLA-A*25, HLA-A*34, HLA-A*36, HLA-A*43, HLA-A*66, HLA-A*69 and HLA-A*74, this frequency was less than 1 %. The frequency of rare HLA-A allelic groups, including HLA-A*34, HLA-A*69 and HLA-A*74, showed a significant difference in the ethnic groups (p-value less than 0.05).
