ارزیابی پیوند همزمان سلولهای بنیادی مزانشیمی انسانی و سلولهای CD34+ خون بند ناف به موش Balb/c اشعه دیده با روش سنجش کلونی طحال CFU-S

<strong><p align="justify"> چکید ه </p><p align="justify"> <strong><i> سابقه و هدف </i></strong></p><p align="justify"> <strong> خون بند ناف حاوی تعداد زیادی سلولهای پیشساز ابتدایی است که برای پیوند بالینی استفاده میشوند ولی درصد موفقیت پیوند این سلولهای بنیادی پایین است. لذا در این مطالعه سلولهای </strong><strong>CD34<sup>+</sup> </strong><strong>همزمان با سلولهای بنیادی مزانشیمی </strong><strong>(MSC) </strong><strong>به موشهای </strong><strong>Balb/c </strong><strong>اشعه دیده، پیوند زده شد و نتایج به صورت شمارش تعداد کلون در سطح طحال ارزیابی گردید. </strong></p><p align="justify"> <strong><i> مواد وروشها </i></strong></p><p align="justify"> <strong> مطالعه انجام شده از نوع تجربی بود. نمونهگیری به روش تصادفی انجام شد. سلولهای </strong><strong>CD34<sup>+</sup> </strong><strong>خون بند ناف انسانی از نمونه خون بند ناف به روش ایمونومگنتیک و </strong><strong>MSC </strong><strong>از مغز استخوان طبیعی انسان جدا شد. </strong><strong>MSC </strong><strong>از نظر مارکرهای سطحی </strong><strong>CD166 </strong><strong>و </strong><strong>CD105 </strong><strong>و درصد خلوص سلولهای </strong><strong>CD34<sup>+</sup> </strong><strong>با فلوسایتومتری ارزیابی شد. سلولهای </strong><strong>CD34<sup>+</sup> </strong><strong>با دوز 10<sup>6</sup>× 2/0 تا 1 به طور همزمان با </strong><strong>MSC </strong><strong>با دوز 10<sup>6</sup>×25/0 تا 1 به موش 6 تا 8 هفتهای اشعه دیده( </strong><strong>Gy </strong><strong>7) پیوند شد. پس از گذشت 11 روز، تعداد کلونها در سطح طحال شمارش و به روش هماتوکسیلین و ائوزین </strong><strong>(H&E) </strong><strong>رنگآمیزی شد. جهت ارزیابی وجود سلولهای </strong><strong>CD34<sup>+</sup> </strong><strong>انسانی در کلون موجود در طحال، این سلولها با ذرات آهن </strong><strong>(SPIO) </strong><strong>نشاندار شده و به موش اشعه دیده تزریق شدند. طحال این گروه به روش آبـی پروس رنگآمیزی شد. در آنالیز آماری جهت بررسی تعداد کلون از آزمایش کروسکال ـ والیس با نرمافزار </strong><strong>SPSS </strong><strong>استفاده شد. </strong><strong></strong></p><p align="justify"> <strong><i> یافتهها </i></strong></p><p align="justify"> <strong> ارزیابی فلوسایتومتری، درصد سلولهای تهیه شده از خون بند ناف و مغز استخوان را برای سلولهای بنیادی </strong><strong>CD34<sup>+</sup> </strong><strong>حدود 90% و </strong><strong>MSC </strong><strong>را بیش از 96% نشان داد و 100 درصد سلولها زنده بودند. تعداد کلونهای طحال در پیوند همزمان دوزهای 10<sup>5</sup>×3 و 2 </strong><strong>CD34<sup>+</sup> </strong><strong>و دوزهای 10<sup>6</sup>×1 و 5/0 </strong><strong>MSC </strong><strong>به طور معنیدار در مقایسه با سایر گروهها افزایش یافت(01/0 </strong><strong>p< </strong><strong>). گرانولهای آهن با رنگآمیزی آبی پروس در کلونهای طحال مشاهده شد. </strong></p><p align="justify"> <strong><i> نتیجه گیری </i></strong></p><p align="justify"> <strong> سلولهای بنیادی مزانشیمی باعث تسریع پیوند سلولهای </strong><strong>CD34<sup>+</sup> </strong><strong>میشود و استفاده از پیوند همزمان سلولهای بنیادی خون بند ناف به همراه سلولهای بنیادی مزانشیمی میتواند موفقیت پیوند سلولهای بنیادی را افزایش دهد. </strong></p><p align="justify"> <strong><i> کلمات کلیدی </i>: سلولهای بنیادی، پیوند همزمان، خون بند ناف، سلولهای مزانشیمی </strong></p></strong>
<p> <strong></strong></p><p> <strong>Abstract</strong> </p><p> <i><strong>Background and Objectives</strong></i></p><p> Umbilical cord blood (UCB) contains a high number of primitive progenitor cells for transplantation. However, the rate of UCB CD34<sup>+</sup> stem cell engraftment is low. In this study we examined the effect of human MSCs (mesenchymal stem cell) on engraftment of human UCB-derived CD34<sup>+</sup> cells in irradiated Balb/c mice. </p><p> </p><p> <i><strong>Materials and Methods</strong></i></p><p> Human UCB CD34<sup>+</sup> cells were obtained from full-term normal deliveries by immunomagnetic techniques and MSCs were isolated by a standard methodology from bone marrow aspirates. Isolated MSCs were evaluated for cell-surface antigens of CD166 and CD105 by flowcytometry. The absolute count of CD34<sup>+</sup> UCB stem cells was also determined by flowcytometry. Irradiated (7 Gy) Balb/c mice were transplanted intravenously with 0.2 to 1.0 × 10<sup>6</sup> human UCB CD34<sup>+</sup> cells in the presence or absence of 0.25 to 1 × 10<sup>6</sup> human bone marrow-derived MSCs. The mice in every group on day 11 after transplantation were killed and their spleen dissected. In every group, colony assay and H and E staining were performed. To make sure of the presence of human stem cells in spleen colonies, UCB CD34<sup>+</sup> cells were labeled with super paramagnetic iron oxide (SPIO) before transplantation. The colonies in spleen then underwent Prussian blue staining. In the statistical analyis, Kruskal-Wallis test using SPSS software was employed to determine the number of colonies. </p><p> </p><p> <i><strong>Results</strong></i></p><p> Flowcytometry assay showed that after purification, 90% of the cells were CD34<sup>+</sup> and 96% marker positive for MSCs. Viablity of cells was 100%<strong>.</strong> Cotransplantation of low doses of UCB CD34<sup>+</sup> cells (0.2 and 0.3 × 10<sup>6</sup>) and MSC (0.5 and 1 × 10<sup>6</sup>) resulted in a significant increase of the number of colony forming units spleen, in comparison with the engraftment of UCB CD34<sup>+</sup> stem cells without MSC after 11 days (p<0.01). Prussian blue staining showed the Fe<sup>+2</sup> granules in UCB stem cells. This indicated that cells in the colonies were engrafted UCB CD34<sup>+ </sup>stem cells. </p><p> </p><p> <i><strong>Conclusions</strong></i></p><p> The results showed that cotransplantation of MSC with UCB CD34<sup>+ </sup>cells promotes engraftment of UCB CD34<sup>+ </sup>cells. </p><p> <i><strong>�</strong></i></p><p> <i><strong> Key words : </strong></i><strong></strong>Stem cell, Cotransplantation, Umbilical cord blood, Mesenchymal stem cell </p>
